Abstract :
This review synthesizes the current landscape of gold nanoparticle (GNP)-assisted hyperthermia for cancer treatment, examining established methodologies, persistent challenges, and promising future directions. We delve into the performance of GNPs in both in vitro and in vivo preclinical models, underscoring their efficacy in enhancing cancer cell death and inducing tumor regression through photothermal effects. The review highlights current approaches, including the progression of GNP-based nanoshells into clinical trials, and explores their synergistic potential when combined with conventional therapies like chemotherapy, radiotherapy, immunotherapy, and gene therapy. Concurrently, we address critical aspects of safety, toxicity, and the hurdles associated with clinical translation. Finally, this review identifies key challenges, such as achieving uniform tumor temperature distribution and overcoming light penetration limitations, while spotlighting innovative future directions, including the utilization of near-infrared II (NIR-II) window agents and hybrid nanoplatforms, to advance GNP-assisted hyperthermia as a robust clinical strategy.
